The pair would ultimately become enemies after the high council bestowed Pax with the title of Prime, choosing his idea of peaceful reform over Megatronus's violent one. An enraged Megatronus, now Megatron, would wage war on the higher classes of Cybertron, prompting the newly appointed Optimus Prime to form the Autobots. In this new position, Prime would command the Autobots throughout the war on Cybertron, during the course of which Megatron proceeds to infect the core of Cybertron, and by extension Primus. Optimus takes the mission of attempting heal the infected core with hopes of saving Cybertron. However it becomes clear that Cybertron is beyond repair, and Primus must shut down to heal, the process leaving cybertron uninhabitable for millenia. Primus, however bestows upon Optimus the Matrix of Leadership before shutting down, making Optimus the de facto leader of the autobots, elevating him to the rank of Prime.The moment was short lived, as Megatron flew in to steal the staff and the Knights arrived to slay Optimus for serving "Quintessa the Deceiver". While Optimus offered no resistance, viewing his execution as proper atonement, Cade managed to activate the Talisman and call off the Knights. Optimus then allied with the Knights and they flew off in the Knight Ship to confront Megatron and Quintessa on Cybertron. Arriving on Dragonstorm, Optimus finally destroyed Infernocus by decapitating all six Infernocons in a single mighty swing of his sword, reminding them that he was Optimus Prime.

Optimus then reunited with the humans and other Autobots and declared that they would never betray Earth, which is their new home with Cybertron destroyed. During the war, Optimus encountered Skyquake at the Battle of Technahar, when the Decepticon was sent by Megatron to kill him. After Cybertron was abandoned, Optimus Prime's elite team of Autobots eventually made their way to Earth, where they allied themselves with the United States government. The Autobots were assigned William Fowler as their liaison, and provided an abandoned missile silo (known as Autobot Outpost Omega 1) as a base in exchange for keeping the Decepticon threat contained. Optimus then sent a message into space, calling any Autobot survivors to Earth. Once a simple data clerk, Optimus Prime was chosen, to his surprise, to become the leader of the Autobots. Optimus Prime believes that freedom is the right of all sentient beings. He uses his strength and bravery to rally his heroic Autobots and wage their battle against the evil forces of the Decepticons Master Swordsman: Optimus is skilled in using multiple swords, specifically his energon swords until they were destroyed by Galvatron, and claimed the Sword of Judgement from one of the Knights Temenos’s armoury. Master Combatant: Optimus is a master hand-to-hand combatant, having extensive levels of knowledge of melee combat as well as being extremely experienced in combat operations. Following Optimus and the Autobots' arrival on Earth, and the subsequent battle of Mission City and the destruction of Megatron and the AllSpark.Flight: As seen in Transformers: Age Of Extinction, Optimus is able to fly, and can also withstand extreme temperatures in space. Optimus Primal tells Optimus Prime that he was named after him (Optimus Prime) this shows that Optimus Prime is older than Optimus Primal.
